297 - Expanding Methods for Evaluating Insecticide Resistance at the Northeast Center for Vector-Borne Diseases TEC

Abstract: The Northeast Regional Center for Excellence in Vector-Borne Diseases (NEVBD-TEC) Pesticide Resistance Monitoring Program was established in 2019 to assess resistance to insecticides in mosquitoes collected from an area covering 13 Northeastern/Mid-Atlantic states and the District of Columbia. Our central focus is to provide resistance testing for regional partners who collect and ship Culex pipiens and Aedes albopictus for testing using the CDC bottle Bioassay or larval cup assay. We see trends of increasing resistance to methoprene and pyrethroids in Cx. pipiens.

A significant gap exists in available diagnostics to assess other important mosquito species that are targets for control in our region, including Culiseta melanura and Culex restuans. To address this gap, NEVBD-TEC developed baseline susceptibility criteria for Cs. melanura against the most utilized active ingredients for adult control. Additionally, with the help of our regional partner agencies, we compared mortality between Cx. pipiens and Cx. restuans to better understand Cx. restuans susceptibility. This work revealed consistently higher mortality in Cx. restuans when compared to Cx. pipiens.

Our program also focuses on development and optimization of resistance assays. We developed a topical assay that uses fewer mosquitoes than other approaches, potentially making resistance testing more accessible and sustainable for regional programs. Additionally, we evaluated the use of formulated products in a bottle-based assay by comparing them to the technical grade CDC Bottle Bioassay. We observed higher mortality with formulated products in Aedes aegypti and Cx. pipiens to Zenivex E20, DeltaGard and Fyfanon ULV. This indicates that using formulated products could mask resistance to the active ingredient and underscores the need for a distinct bench assay using formulated products as a first line method of evaluating product efficacy.
